Tweelingresultaten van niet-coderende RNA’s (ncRNA’s) in de meeste vormen van kanker Stamcelbiologie
De identificatie van de meeste kankerstamcellen (CSC’s) als initiatoren van carcinogenese heeft een revolutie teweeggebracht in de periode van de meeste kankeranalyses en ons begrip van de keuzes voor het verhelpen van ziekten. Extra CSC-opties, samen met zelfvernieuwing en migrerende en invasieve mogelijkheden, hebben deze cellen extra gerechtvaardigd als vermeende diagnostische, prognostische en therapeutische doelen.
Gezien de CSC-plasticiteit is de identificatie van CSC-gerelateerde biomarkers een zware last geweest bij CSC- karakterisering en therapeutische concentratie. In de afgelopen lange tijd heeft een overtuigende hoeveelheid bewijs cruciale regelgevende mogelijkheden van niet-coderende RNA’s (ncRNA’s) op de unieke opties van CSC’s aangetoond.
We weten nu dat ncRNA’s kunnen binnendringen in signaalroutes, erg belangrijk voor het onderhoud van het CSC-fenotype, vergelijkbaar met Notch, Wnt en Hedgehog. Hier hebben we het over de veelzijdige bijdrage van microRNA’s (miRNA’s), lange niet-coderende RNA’s (lncRNA’s) en ronde RNA’s (circRNA’s) , als consultant ncRNA-cursussen, bij het ondersteunen van de CSC-achtige eigenschappen, naast de onderliggende moleculaire mechanismen van hun beweging in gevarieerde CSC-variëteiten. We praten verder over het gebruik van CSC-gerelateerde ncRNA’s als vermeende biomarkers van buitensporige diagnostische, prognostische en therapeutische waarde.
Electrospun polyurethaan / poly (ɛ-caprolacton) nanovezels bevorderde de bijlage en vooruitgang van de menselijke endotheliale cellen in statische en dynamische aangepaste voorwaarden
- Bij deze studie werd de angiogene functionaliteit van menselijke endotheelcellen bestudeerd na 72 uur op de grond van polyurethaan- polycaprolacton (PU/PCL) steigers te zijn uitgeplaat . In deze studie zijn cellen ingedeeld in 5 totaal verschillende groepen, samen met PU, PU/PCL (2:1), PU/PCL (1:1); PU/PCL (1:2); en PCL.
- Info onthulde dat de PU/PCL (2:1) samenstelling de volgende modulus en breekpunt had in tegenstelling tot de andere groepen (p<0,05). In vergelijking met de andere groepen had de PU/PCL-steiger met een molverhouding van twee:1 een lagere contacthoek θ en een hogere trekspanning (p<0,05).
- De indicatieve meting van de PU-nanovezels werd verlaagd na de toevoeging van PCL (p<0,05). Op basis van onze informatie veroorzaakte de gewoonte van endotheelcellen op basis van PU/PCL (2:1) geen nitrosatieve stress en cytotoxische resultaten onder statische omstandigheden in vergelijking met cellen die waren uitgeplaat op een conventionele plastic vloer (p>0,05).
- Op basis van informatie uit de statische stand van zaken, hebben we een buisvormige PU/PCL (2:1) assemblage gefabriceerd voor zes dagen durende dynamische cel op maat gemaakte inside-loop air-lift bioreactoren. Scanning-elektronenmicroscopie bevestigde de hechting van endotheelcellen aan de luminale vloer van de PU/PCL-steiger. Cellen zijn afgeplat en uitgelijnd onder de aangepaste medium beweging. Immunofluorescentiebeeldvorming bevestigde de hechting van cellen aan de luminale vloer, aangegeven door blauwe kernen op de luminale vloer.
- Deze informatie toonde aan dat de uitrusting van PU/PCL-substraat de trein van endotheelcellen zou kunnen stimuleren onder statische en dynamische omstandigheden.

Gedurende het laatste decennium heeft de knowhow op het gebied van driedimensionale (3D) celgebruik verschillende nieuwsgierigheid opgedaan die toe te schrijven is aan het potentieel om de in vivo groep en micro-omgeving van in vitro gekweekte meeste kankercellen beter te recapituleren.
Het is veelbetekenend dat 3D-tumormodes verschillende totaal verschillende eigenschappen hebben aangetoond in tegenstelling tot standaard tweedimensionale (2D) culturen en een opvallende hyperlink hebben aangebracht tussen de laatste en dierproeven.
Eigenlijk kenmerken 3D-celculturen een nuttig platform voor de identificatie van de natuurlijke keuzes van de meeste kankercellen, samen met voor de screening van nieuwe antitumormakelaars. Temperatuurgevoelige methylcellulose-hyaluronhydrogel als een 3D-celcultuurmatrix
- Deze studie onderzocht de uitrusting van een temperatuurgevoelige methylcellulose-hyaluronzuur (MC-HA) hydrogel om de voortgang van 3D-cellen in vitro te helpen. Het voorbereidende werk concentreerde zich op de voorbereiding van hydrogels voor 3D-gebruik , aangenomen door onderzoeken naar de natuurlijke compatibiliteit van hydrogel-elementen en optimalisatie van de aangepaste celomgeving.
- Evaluatie van de levensvatbaarheid en proliferatie van HCT116-cellen die in de MC-HA-hydrogel waren gekweekt, werd gebruikt om de combo-samenstelling te controleren om een hydrogel te ontwerpen met optimale eigenschappen om de celvooruitgang te helpen.
- Twee zeer belangrijke factoren met betrekking tot de bruikbaarheid van de voorgestelde polymere matrix in 3D-celgebruik zijn aangetoond: i) 3D-gekweekte celaggregaten zullen waarschijnlijk uit de matrix worden gelanceerd/teruggewonnen door middel van een fragiel verloop dat de levensvatbaarheid van de cellen kan verdedigen, en ii) de hydrogelmatrix kan worden gebruikt in plaatformaat met 96 putjes als een dagelijkse techniek die wordt gebruikt bij in vitro weefselspecifieke onderzoeken.
- Het werk vanwege deze realiteit onthult dat MC-HA-hydrogels potentieel bieden voor in vitro 3D-celgebruik als goedkope en goed gedefinieerde alternatieve keuzes voor van dieren afgeleide of geavanceerde synthetische strategieën.
Primordiale kiemcellen van muizen: in vitro aangepaste en conversie naar pluripotente stamcelsporen
Primordiale kiemcellen (PGC’s) zijn de embryonale voorlopers van de gameten. Ongeacht een zeer lange studietijd, in vitro gebruik van PGC’s blijft een belangrijk probleem en heeft vooraf vertrouwd op ongedefinieerde elementen die verwant zijn aan serum en feeders.
Met name PGC’s die voor langere intervallen zijn gekweekt, behouden hun afstammings- ID niet, maar dragen in plaats daarvan conversie naar variëteit pluripotente stamcelstammen die vaak embryonale kiemcellen (EG) worden genoemd als reactie op LIF / STAT3-signalering. Juist hier rapporteren we alle gevestigde en nieuwe methoden om EG-cellen af te leiden, in heel veel verschillende omstandigheden.
We zijn van mening dat grote bezorgdheid over de voortgang van fibroblasten eenvoudigweg niet vereist is voor EG-celconversie. We houden rekening met de stappen die in ons laboratorium zijn genomen om systematisch geavanceerde elementen weg te nemen en een volledig geschetst protocol te regelen dat een aangename conversie van geïsoleerde PGC’s mogelijk maakt naar pluripotente EG-cellen mogelijk maakt.
Daarnaast presenteren we dat PGC’s op maat kunnen hechten en zich vermenigvuldigen zonder de hulp van voedingscellen of serum. Het zal met succes nieuwe benaderingen adviseren om op korte termijn gebruik van PGC’s in geschetste omstandigheden tot stand te brengen.
